Narrative:

Company dispatcher had changed company standard route from dtw to myr to a new route. During my cockpit preflight procedures, a new hire gate agent handed me a passenger and load manifest that did not correspond to the passenger or bags that were on the aircraft. I asked the gate agent why the numbers were so off from what we had on the aircraft. She said she was new and did not know how to fill out the paperwork for the load manifest. I then had to request another ramp agent to find the true numbers of passenger and bags. This caused me to miss the new routing on the flight release. ATC noticed that I had made a wrong turn and question my filed route. The controller asked if dtw clearance delivery cleared me as filed. I said yes. ATC reclred me to the original route that was on flight release.
