Narrative:
I was training on local control. Aircraft Y was lined up at RY3 full length; and aircraft X was cleared for takeoff from RY3 at charlie intersection. Aircraft X was issued a left turn heading 250; and shipped to departure. Aircraft Y was cleared for takeoff on the departure procedure. Aircraft X did not make a left turn; so we got a turn for aircraft Y to avoid losing separation. Upon audio review it was determined there was no read back of the heading 250. Fortunately the timely action of getting a turn for aircraft Y avoided a loss.I will be more diligent about ensuring readbacks for critical instructions while training.

Title: GEG Tower Controller reported of a near loss of separation due to the a departure not turning as instructed.
Narrative: I was training on Local Control. Aircraft Y was lined up at RY3 full length; and Aircraft X was cleared for takeoff from RY3 at Charlie intersection. Aircraft X was issued a left turn heading 250; and shipped to departure. Aircraft Y was cleared for takeoff on the departure procedure.
