Narrative:

I was working local east and west combined at the local east position. Day VMC conditions with visual approaches to 5R. Runway 5L was closed. Ground control had apreq'd 3 runway 14 departures for southbound aircraft which I approved. A B737 was initially instructed to luaw on runway 5R and then instructed to fly runway heading and take off clearance was issued. An E145 was the next aircraft to depart which I instructed to luaw runway 5R. As I was scanning down the runway; I noticed a C208 enter runway 5R/23L from the approach end of 23L...immediately I tried to stop the B737 and cancel his take off clearance; however it was too late for him to abort and the B737 overflew the C208 by about 500-600 ft. The C208 had made an abrupt maneuver and departed the side of the runway into the grass; then taxied back onto the runway and exited at A7. All procedures were followed. Asde-X may have alerted me to the situation a second earlier.

Title: RDU Controller described a runway incursion event that resulted in a departure over flying an aircraft that had entered the runway without authorization.
